1.Development and neuronal application of G-CaMP-type genetically encoded Ca2+ indicators with high-sensitivity and high-responsivity
神経の活動を可視化することができる最新のツール(カルシウムインディケーターGCaMP)について理解を深め、議論を行なった。(話題提供:大倉、中井)
2.Real-time visualization of neuronal activity during perception
GCaMPを用いたイメージングにより、外界の様子が視覚を通じて、どのように脳に投影されるかについて理解を深め、議論を行なった。(話題提供:武藤、川上)

GCaMPを用いたイメージングにより、脳における匂い刺激情報処理について理解を深め、議論を行なった。(話題提供:Yaksi)

動物の運動における聴覚刺激と、視覚刺激の相互作用について神経回路レベルでの理解を深め、議論を行なった。(話題提供:Du)

運動を抑制的に制御するグリシン作動性シナプスの形成機構について理解を深め、議論を行なった。(話題提供:平田)
第1回研究会における上述の議論を通じて、
(1)ひとつの細胞(神経細胞およびその他の細胞)、ひとつの神経回路の活動を検出する、およびそれらを操作するための最先端の技術についての知識を共有し議論することにより、それら技術の現時点における問題点、将来の可能性、必要な改良等についての研究を推進することができた。
(2)また、それら細胞(神経細胞およびその他の細胞)の活性を動物個体の行動(その他の細胞の場合は活性の総和としての器官形成)と対応づけるための方法論について同様に議論し、研究を推進することができた。
これらの成果をもとに次年度以降も、脊椎動物が示す一見複雑にみえる行動を規定する基本的な神経回路はどのようなものか、それらを基にしてどのように神経ネットワークが構築され機能しているのか、を明らかにするための研究を継続して発展させていく。

2. Real-time visualization of neuronal activity during perception

We discussed visualization of neuronal activity in the brain during visual perception.
3. Studying neural circuit function in zebrafish habenula
Emre Yaksi(Neuro-Electronics Research Flanders、Leuven、Belgium)
We discussed how the brain (habenula) processes olfactory information.
4. Neural circuit basis of olfactory behavior in zebrafish

We discussed genome engineering by homologous recombination in zebrafish.
9. Exploring roles of ADAM proteases in development using zebrafish

We discussed the roles of the ADAM protease family in the vertebrate organogenesis.
10. In vivo imaging of dendritic refinement of barrel cells in the neonatal mouse cortex
Takuji Iwasato (National Institute of Genetics)
We discussed the mechanism of synapse formation in the mouse barrel cortex.
11. Optogenetic control of glia and mind
Ko Matsui (Tohoku Univ)
We discussed optogenetic studies of the brain function using the channelrhodopsin.
12. Functional analysis of locomotor circuits in the spinal cord and brainstem
Shin-ichi Higashijima (OIIB)
We discussed the study of specific neuronal circuit that controls locomotion by electrophysiological and optogenetic approaches.
13. Dopaminergic gating of visual flow to auditory escape circuit via inhibition

We discussed interaction of visual and auditory inputs during animal behaviors.
14. Formation and function of glycinergic synapse in zebrafish
Hiromi Hirata (National Institute of Genetics)
We discussed formation of glycinergic inhibitory synapses that control animal locomotion.
Thus, we could advance the study to get insights into recent development and future perspective of tools and methodologies to visualize and manipulate the activity of single neuronal circuits and to understand how these methodologies should be used to relate the activity of neuronal circuits to complex animal behaviors.
2013年度:
2013年度は、海外からの講演者を交えた研究会を2月27−28日に開催した。海外研究者招聘の経費等を考慮して年1回の会議の開催とした。
今回の会議では、イメージング手法、オプトジェネティクス手法を開発、あるいはそれらを駆使してモデル動物における脳神経回路機能研究の分野で活躍している国内外の研究者が、講演・話題提供を行なった。海外から3名の研究者を招聘し、以下のテーマで話題提供を行ない、それらについて議論・考察した。

これらを通じて、カルシウムセンサー等最新のイメージング技術、チャネルロドプシン等の最新の光遺伝学(オプトジェネティクス)の技術、Crisper/CAS9、Talen、Transposon等最新の遺伝子工学技術について洞察を深め、神経科学研究、脳機能研究から新しい概念を生み出すためのアプローチについて研究した。 |
